INFLUENCE OF SELECTED POLYPHENOLS ON ANTIOXIDANT ENZYMES ACTIVITY AND EXPRESSION OF BAX AND BCL-2 GENES IN CACO-2 CELLS EXPOSED ON OXIDATIVE STRESS

Abstract: The main purpose was to study the influence of antioxidants on human colon adenocarcinoma cells Caco-2 exposed to H2O2-induced oxidative injury. Caco-2 cells were exposed twice to 400 or 800 µM of H2O2, with 6 hours of interval between the applications. To assess the impact of quercetin (Qw), chlorogenic acid (ChlA) and epigallocatechin (EGC), cells were pre-treated with the antioxidants solutions (100 or 200 µM) for 24 h prior to H2O2 treatment. The expression of mRNA for bax and bcl2 genes and the activity of intracellular antioxidant enzymes (catalase /CAT/, glutathione peroxidase /GPx/) in Caco-2 cells were estimated 24 and 48 hours after exposition to the stress agent. We demonstrated that pretreatment with analyzed antioxidants can activate anti-apoptotic signal in cells exposed on oxidative injury and prevent their apoptosis. On the basis of the results we found that protective effect of antioxidants is independent on antioxidant enzymes activity.
